In the Kraut lab, we study the proteasome, a macromolecular machine inside all eukaryotic cells responsible for unfolding and degrading proteins. We are specifically interested in factors that influence proteasomal processivity, or the tendency of the proteasome to either degrade or release its substrates.
We use mutant and wild-type yeast proteasome, purified substrates and enzyme kinetics to probe proteasomal unfolding ability.
Students in the Kraut Lab learn molecular biology, yeast genetics, protein purification, and numerous in vitro enzyme assays.
